A method for calculating the curvature of each point on the cell image surface based on shadow restoration technology is proposed. The boundary contour extraction technology is used to separate the red blood cell brightness image obtained by the scanning electron microscope (SEM). The region growth of a single cell starts from the center point obtained by tracking. The brightness change information of each pixel determines the corresponding height field depth. The shadow restoration technology can convert this changing brightness into different depths, thereby obtaining the three-dimensional data points of the pixel. The surface fitting of the three-dimensional points is performed by the least squares method to obtain a piecewise continuous and smooth cell surface, and its shape characteristics are represented by the average curvature and Gaussian curvature.
