Polymerase chain reaction based on dendrimers: PCR based on the interface of dendrimers (PAMAM) was studied. The primers were connected to the surface of PAM2AM molecules through electrostatic interaction. Plasmid pBluescript SK was used as a template for PCR. The PCR products were detected by agarose gel electrophoresis and transmission electron microscopy. The effects of annealing temperature and primer concentration on PCR were studied respectively. The experimental results showed that PCR with primers connected to the surface of PAMAM molecules could be successfully carried out, and within a certain temperature range, PCR was almost unaffected by annealing temperature. PAMAM-DNA polymers were prepared by PCR. This polymer can disperse at high temperatures and compound at low temperatures. PCR based on the surface of PAMAM molecules extends the PCR technology in homogeneous systems to heterogeneous systems, greatly expanding the application range of PCR technology and providing a new method for preparing nanomaterials.
